Robust Stability Analysis and Design Under Consideration of Multiple Feedback Loops in the Tryptophan Regulatory Network of E. coli

چکیده
The tryptophan operon of E. coli represents an important regulatory unit consisting of multiple feedback loops. The role of these loops is crucial to understand the dynamics of tryptophan biosynthesis. We analyze the robust stability of this system by splitting its state into fast (mRNA concentration) and slow (anthranilate synthase and tryptophan concentrations) variables, under consideration of nonlinear uncertainties. In addition, we analyze the role of these feedback loops as key design components, responsible for the physiological performance of this regulatory unit. The range of allowed parameter perturbations and the conditions that ensure the existence of asymptotically stable equilibria for the perturbed system are determined. We also analyze two important alternate regulatory designs for the tryptophan synthesis pathway and derive the corresponding stability conditions.

Multiple feedback loop design in the tryptophan regulatory network of Escherichia coli suggests a paradigm for robust regulation of processes in series.
Biological networks have evolved through adaptation in uncertain environments. Of the different possible design paradigms, some may offer functional advantages over others. These designs can be quantified by the structure of the network resulting from molecular interactions and the parameter values. One may, therefore, like to identify the design motif present in the evolved network that makes ...
متن کاملMultiple feedback loops are key to a robust dynamic performance of tryptophan regulation in Escherichia coli.
Living systems must adapt quickly and stably to uncertain environments. A common theme in cellular regulation is the presence of multiple feedback loops in the network. An example of such a feedback structure is regulation of tryptophan concentration in Escherichia coli. Here, three distinct feedback mechanisms, namely genetic regulation, mRNA attenuation and enzyme inhibition, regulate tryptop...

MOTIVATION Cells continuously reprogram their gene expression network as they move through the cell cycle or sense changes in their environment. In order to understand the regulation of cells, time series expression profiles provide a more complete picture than single time point expression profiles. Few analysis techniques, however, are well suited to modelling such time series data. RESULTS ...
